What is PICSI
What is PICSI?
PICSI (Physiological Intracytoplasmic Sperm Injection) is a refinement of ICSI used in assisted reproduction. Instead of selecting sperm by appearance alone, PICSI uses hyaluronic acid — a molecule resembling the substance naturally found in the cells surrounding the egg — to identify mature sperm.
Only a mature, biochemically competent sperm carries the receptors needed to bind hyaluronic acid. Studies suggest that sperm which bind in this way tend to have better morphology, lower chromosomal abnormalities and improved DNA integrity, which is why PICSI is used to improve sperm selection in selected cases.

Step by Step
How PICSI is performed
A physiological selection step is added to the standard ICSI workflow.

Hyaluronan dish
A dish is coated with hyaluronan, the substance found in the cumulus cells surrounding the egg.

Semen processing
The sample is processed to separate sperm from the seminal fluid.

Sperm binding
The processed sperm are placed on the hyaluronan-coated dish; mature, healthy sperm bind via their specific receptors.

Identification
Under the microscope, the bound sperm — those attached to the hyaluronan — are identified.

Injection
Each selected sperm is picked up with a fine needle and injected directly into the egg cytoplasm, as in ICSI.
